Glutaminase, an amidohydrolase enzyme that catalyzes the hydrolysis of L-glutamine to L-glutamic acid and ammonia, has emerged as a potential anticancer agent due to its ability to deprive cancer cells of glutamine—an essential nutrient for their rapid proliferation. This study explores the isolation, screening, and evaluation of bacterial strains for the production of glutaminase with potential anticancer properties. Soil samples from diverse habitats were collected and processed using serial dilution and selective media techniques to isolate glutaminase-producing bacteria. Preliminary screening was performed using phenol red-glutamine agar, followed by quantitative enzymatic assays to assess glutaminase activity. Crude enzyme extracts were subjected to in vitro cytotoxicity tests against selected human cancer cell lines to assess anticancer potential. Several bacterial strains demonstrated significant glutaminase production with notable cytotoxic effects, indicating their promise as bioresources for enzyme-based cancer therapy. This research contributes to the identification of novel microbial strains capable of producing therapeutic enzymes and provides a foundation for future biochemical and pharmacological investigations of glutaminase as a natural anticancer agent.
