Importance of Pyroptosis in Human Diseases

Abstract:

The importance of Pyroptosis has drawn many attentions to scientists and medical personnel as it has been found to be associated with innate immunity and many human diseases. This is an inflammatory programmed cell death with some important differences from other cell death like Apoptosis, Necroptosis, Necrosis and others. The difference between some important cell death has been discussed. As pyroptosis occurs generally after some infections with pathogens, so it is sometimes called Primary Programmed Cell death. It is genetically regulated and highly inflammatory type of cell death. The pyroptotic death or its pathway is involved with many components, such as, Inflammasome, Caspases and its substrate Gasdermin and these are closely related with many important diseases including cancer. In this review, the role of pyroptosis in the progression of tumour or cancer progression has been discussed. The dual function of pyroptosis has also been mentioned. On one side, its pro-inflammatory response gives immune cells to fight pathogens and on the other side the overabundance of pyroptosis (due to some disrupted pathways) results in the release of cytokines leading to cytokine storm inducing lethality. The role of pyroptosis in cardiovascular and cancer diseases has been discussed. As the important components of pyroptosis, inflammasomes and gasdermin can promote cancer growth as well as it can also suppress cancer growth, so it has a great potential in its therapeutic application that also has been written.
